报告摘要:
Single-molecule manipulation technologies can apply various external mechanical constraints to a single biological molecule and measure its mechanical responses. The mechanical response of a molecule is sensitive to the interactions with its binding partners (other proteins or drugs), making such measurements a powerful approach in quantifying molecular interactions at an ultra sensitive single-molecule level. In the last ten years, our lab has developed a set of highly efficient approaches that have allowed us to manipulate various DNA structures (dsDNA, ssDNA, G-quadruplex, etc), protein structures (folded domains and unfolded peptide chain), and to quantify biomedically crucial interactions between each structure and its partners. In this talk I will present the general principles of mechanical manipulations, the details of how each structure is created and manipulated in our lab, the characteristic mechanical response of each structure. Further, I will demonstrate how interactions can be detected using one short example for each structure. Finally, I will focus on one recent study of force-dependent alpha-catenin – vinculin interaction to demonstrate how such technologies can make biological discovery.
报告人简介:
Jie Yan is currently a Dean’s Chair associate professor in the department of physics and a principal investigator in the mechanobiology institute, National University of Singapore. His laboratory has been focused on understanding micromechanics of DNA, protein, and their interactions, at a single-molecule level, to address important questions covering a broad spectrum of areas such as DNA organization and gene regulation, DNA damage repair, and mechanosensing of cells.
